These two have actually interacted before. And I genuinely believe that Penn and Teller would work a crowd better than Al, cause they can sing and dance (as shown in the video above), as well as being able to do magic tricks. And, naturally, they are well versed in comedy.

Weird Al is also well versed in Comedy.  He's the elder statesman of Comedy.

Weird Al can sing and dance I'd say better than Penn and Teller.

Magic tricks... not sure.

Damn, this is so even it's crazy.

My thoughts on how it plays out: Both can do the comedy opener well, I don't think either has and edge there. Dance Finale is even too.

The difference comes in the song and the magic trick, and it's a game of margins. Penn and Teller will absolutely dominate the magic trick by a very wide margin. Weird Al will do better in the song, but as Macklemore presented above Penn and Teller can compete there, so the margin is smaller.

Overall, the longest headliners in Vegas, Penn and Teller, win.

Penn and Teller have been in showbiz forever

Heh, so has Weird Al. Both acts go back to the 1970's.

Judging from that great old video, while I wasn't impressed with Penn's singing, the rest of his and Teller's act matches up pretty well with Al's.

I'm honestly torn on this one.

In all seriousness, I think this might be a draw, but I'm leaning towards Al. Weird Al takes singing commandingly, Penn and Teller are better magicians, comedy could go to either depending on the crowd, and dancing I just feel that Weird Al would be marginally better at.

I think calling the song/dance closer than it ought to be. P/T are awesome, but they aren’t performing a song on weird al’s level. Assuming he knows he’s prepping for them, Weird Al probably writes some crazy parody on Penn and Teller, whipping the crowd onto his side. The comedy is probably the closest part, idk, probably depends on whether you like the stand up P/T do or the theatrics of Al.

i feel like Al is going to win the dance finale. Literally being one of the Earth’s most accomplished accordion players, that’s probably a little better Than what P/T could do.
