I feel like that's selling Kool Aid Man a bit short. He's iconic, sells his product well, and has lasted a long time.

All that said, I do feel Flo might have the advantage just because she's a more modern advertising icon. It would feel like pandering to give the Kool Aid Man a smart phone and have him try to sell it

If Amazon wants the advertising to really stick in people's minds then Kool Aid Man is the way to go. A commercial starring Flo would just get people thinking the actress now works for Amazon and then immediately go about their day. On the other hand, a commercial where people are discussing how cool it is to get things off Amazon and then complaining about their current phones only to have the Kool-Aid man burst through the wall to tell them about the Fire phone, that would get people talking. The out of left field pick, the absurdity, and the pull of nostalgia all working in tandem to get people talking about this new phone. 

"Oh Yeahh!!!"
