So Samus would have a wider variety of skills that would let her attack from multiple angles. Assuming they're put into Cuphead as is, Luigi would only really have fireflowers and punches and kicks. The punches we could say function like the spreadshot and the fireflower the lobber, that severely restricts Luigi's attack options. Samus, on the other hand, essentially has the charge shot by default, as well as various other types of weapons that could fill in for most of the Cuphead attacks, letting her counter many bosses that Luigi would really struggle with. In fact, Cagney, the boss in the picture, would be really hard to beat with Luigi's loadout.

If we assume they get access to Cuphead weapons, Samus is still far more skilled with this type of thing than Luigi, and her armor undoubtedly would protect her from some of the attacks

We have to note that Luigi has some skills that may be applicable here.  Potentially jumping onto an enemy to defeat them and also picking up objects and even enemies and trowing them.

Luigi is more martial than Samus.  However, Cuphead is a game that favors ranged attacks.

Even then, I doubt that. Samus is just as agile as Luigi, if not more so. Her ability to turn into a ball (literally impossible for Luigi if I had to wager), coupled with the fact that she was raised by birds to be just as agile and light as them should make her a nice match up for Luigi. This isn't accounting the fact that Cuphead is basically a shooter game, which is definitely in Samus' house moreso than Luigi's.
